Centrifuge

A centrifuge is based on the principle of a centrifugal force and is used to separate substances based on the density. The heavier materials are pushed to the outside of the vessel. This force is generated by spinning the rotor of the centrifuge at very high speeds. 

There are different types of centrifugation like

  • Differential centrifugation: This is used to separate particles of different densities. The larger particles being denser will settle faster.

  • Rate zonal centrifugation: In this type of centrifugation, the sample is layered over a density gradient, which prevents the heavier particles from contaminating the lighter ones.

  • Isopycnic centrifugation: The components of a sample are separated due to the density gradient formed during high-speed centrifugation. When the density of the particle is the same as the density of the surrounding medium, it stops settling.  

Microcentrifuge, benchtop centrifuge, vacuum centrifuge, clinical centrifuge, and ultracentrifuges are the different types of centrifuges that are widely available. A microcentrifuge or a microfuge is used to spin small amounts (less than 2ml) of the sample. These are relevant for the separation of biological samples (eg. Nucleic acids). They are usually benchtop models with refrigerated or ventilated variants. Ultracentrifuge is optimized to achieve very high speeds, in the range of 9800 km/s2. There are two types of ultracentrifuges, preparative and analytical. A clinical centrifuge is used in hospitals and research centres for the separation of serum, plasma or blood samples.
