Chromatography Columns

Chromatography is used to separate and isolate chemical compounds from a mixture. The mixture is allowed to move through a stationary phase and based on the differential absorption rates, the samples separate out. Column chromatography uses a stationary phase packed into a cylindrical tube. Some of the most common stationary phases include silica gel, alumina etc. The stationary phase can be designed to suit ion-exchange, forward or reverse phase chromatography, affinity chromatography etc. There are two methods by which the column is prepared for chromatography the dry method and the wet method. In the dry method the column is filled with the dry stationary phase and then the liquid mobile phase is allowed to wet the stationary phase. In case of the wet methods, the mobile phase and the stationary phase are mixed well and packed into the column. The components of the mixture are separated by retention on the stationary phase due to adsorption. The eluent is collected using an elution buffer.
