10X DNA Loading Buffer is supplied by HiMedia. It is available in the form of a blue coloured solution. The 10X DNA Loading Buffer is composed of Bromophenol Blue, Xylene Cyanol FF, EDTA and glycerol. The EDTA in the loading buffer terminates the reactions containing enzymes that require Mg2+ and Ca2+. Glycerol increases the density of the sample so that it can settle in the well. Addition of one volume of 10X DNA Loading Buffer to nine volumes of DNA sample gives color to the sample as well as increases the sample density, which facilitates the loading onto the well. Furthermore, the solution contains two dyes which migrate in the same direction as the nucleic acids, hence serves as an indicator of the electrophoretic progress. 10X DNA Loading Buffer is composed of 0.21% Bromophenol Blue, 0.21% Xylene Cyanol FF, 0.2M EDTA, pH 8.0 and 50% Glycerol. It is advisable to store this product at 2-8C.

The 10X DNA Loading Buffer is widely used for loading DNA samples into gel electrophoresis wells and tracking migration during electrophoresis. It is provided in a premixed, ready-to-use form. The product is mixed with DNA samples to facilitate the loading of the samples into the wells of horizontal and vertical agarose and polyacrylamide gels. The 10X DNA Loading Buffer is used for analysis of DNA samples including genomic and plasmid DNA, PCR products, restriction fragments etc. on agarose or polyacrylamide gels. In a 0.51.4% agarose gel in 0.5X TBE, xylene cyanol FF migrates at approximately 4kb, bromophenol blue at approximately 300bp.
