It is available in the form of a colourless solution with a pH in the range of 8.2 to 8.4. 10X Transfer Buffer is mainly used to transfer proteins from polyacrylamide gels to nitrocellulose or PVDF (Polyvinylidene fluoride) membranes during Western blotting procedure which uses antibodies to identify specific protein targets bound to the membrane. to transfer proteins onto a membrane (PVDF/Nitrocellulose) for Western blot. This buffer is supplied as a 10X stock. 10X Transfer Buffer is composed of 0.25 M Tris and 1.92 M Glycine and the pH is adjusted to 8.3. It is advisable to store this product at 2-8C.

Western blotting procedure was first introduced by Towbin in 1979 and has become a routine process for protein analysis. The first step of this procedure is to separate the protein samples according to their size by SDS-PAGE.
