High grade Amies Transport Medium w/ Charcoal is supplied by Micromaster. It is available in the form of a grey to black homogenous free flowing powder which has a pH around 7-7.4. This product should be stored between 10-30°C in a tightly closed container. Once opened, it should be properly stored, after tightly capping the bottle. Due to the hygroscopic nature of the product, there could be chances of lump formation which can be avoided by proper storage. Store in dry ventilated area protected from extremes of temperature and sources of ignition.

The pre-requisite of any transport medium is that it should be non-nutritive, semi-solid, and reductive and should be able to hamper self-destructive enzymatic reactions within the cells and in addition, must inhibit toxic oxidation reactions. Amies modified Stuart's Transport Medium by replacing glycerophosphate with an inorganic phosphate buffer and adding charcoal to the medium. This modified medium gave a higher percentage of positive results than the transport medium of Stuart. This medium contains sodium chloride (0.3% concentration) for the optimal preservation of Neisseria gonorrhoeae. Potassium, calcium and magnesium salts maintain osmotic equilibrium by controlling the permeability of bacterial cells. Phosphate buffer maintains pH of the medium. Sodium thioglycolate and agar aid in providing a reduced environment favourable for a wide range of bacteria. Charcoal neutralizes metabolic products toxic to gonococci. Amies Transport Medium provides a reduced environment due to the presence of sodium thioglycollate and small amount of agar. Calcium, magnesium, potassium and sodium salts help the survival of gonococcal cells and also control permeability of bacterial cells. Phosphates buffer the medium.
