This Alpha-amylase test kit is used in clinical biochemistry. The test is used to determine the levels of ammonia in the blood plasma. The test uses the reaction between ammonia, NADH and -ketoglutarate, in the presence of glutamate dehydrogenase (GLDH), to give the products glutamate and NAD+. As the NADH+ gets converted to NAD+, the absorbance reduces and it can be measured spectrophotometrically at 304 nm. This reduction is considered to be a direct indicator of the plasma levels of ammonia.
