The Chromogenic Coliform Agar is a type of medium used to identify and count -glucuronidase-positive Escherichia coli and -D-galactosidase-positive coliform bacteria in water samples through membrane filtration. The medium contains peptone and yeast extract as a source of nitrogen and vitamins, while sodium chloride maintains the osmotic balance and the phosphates act as a buffer. Sodium pyruvate, sorbitol, and tryptophan help in the detection and recovery of the target organisms. Chromogenic Coliform Agar medium uses dual chromogens to differentiate between E. coli and coliforms based on their enzymatic activity. E.coli shows both -D-galactosidase and -glucuronidase activity, which makes them appear dark blue to violet in colour.
