EXTRACTME DNA CLEAN-UP & GEL-OUT KIT, 250 preps supplied by BLIRT is intended to ensure a quick and effective DNA purification after enzymatic reactions and and directly from agarose gels (standard and low-melting point agarose gels run in either a TAE or TBE buffer).. It effectively eliminates nucleases, enzyme inhibitors, detergents, restriction enzymes, polymerases, divalent ions, agarose, ethidium bromide and other contaminants, etc. The purified DNA can be utilized in general downstream applications. The protocol of isolation and buffer formulations was optimized to ensure high efficiency of isolation and purity of pDNA. The process for DNA purification uses spin minicolumns with membranes which bind nucleic acids efficiently and selectively. The 1st isolation step involves CB Buffer is added to DNA sample. This leads to the degradation of proteins and enabling DNA to bind with the columns membrane while in the gel-out protocol DNA fragments is excised from an agarose gel and incubated in GB Buffer, which enables gel fragment solubilization and protein degradation. The binding buffer has a color indicator that allows an easy monitoring of the solutions pH for optimal DNA binding. The two-step washing stage adequately eliminates pollutants and enzyme inhibitors. The elution of purified DNA is done with the help of either a low ionic strength buffer (Elution Buffer) or water (pH 7.09.0). And it may be utilized directly in all downstream applications such as PCR, qPCR, DNA sequencing, enzymatic restriction, DNA ligation, etc. or stored until ready to use.
