GN Broth (IP 2018) is supplied by TM Media. It is used in the enrichment of Shigella species from various products as per Indian pharmacopoeia. GN Broth is used to selectively isolate Shigella species, followed by isolation on XLD Agar, a selective medium. Croft and Miller were able to isolate a greater number of Shigella strains from rectal swabs using this method. Meanwhile, Taylor and Schelhart demonstrated that GN Broth was more effective than selenite enrichment media for isolating Shigella. The medium includes polypeptone peptone which furnishes bacterial growth with amino acids and other nitrogenous substances. The mixture of sodium citrate and sodium deoxycholate acts as an obstacle to Gram-positive and certain Gram-negative bacteria like coliforms. Phosphates play a role in buffering the system while sodium chloride helps maintain osmotic balance.
