sodium bicarbonate. The medium is specially designed by Harry Eagle to meet the specific nutritional requirements of certain subtypes of HeLa cells and normal mammalian fibroblasts. Hank's salt mixture does not require CO2 air mixture as it is developed to equilibrate with air. Therefore cells can be grown in reduce CO2 or CO2 free environment. The medium includes the stabilised dipeptide form of L-glutamine, L-alanyl-L-glutamine. Dipeptide form prevents toxic build -up of ammonia by preventing the intramolecular cyclization reaction. L-alanyl-L-glutamine incorporates with L-alanine to protect the alpha

amino acid release both L-glutamine and

L-alanine for use by the cell by breaking the dipeptide within the cell. The moderate release of L-glutamine removes the need of frequent supply of L-glutamine, thus enables liquid media to be stored at 4C for

longer periods.
