Nigrosin is a high-grade reagent supplied by Micromaster. It is also referred to as acid black 2 sometimes. Nigrosin is available in the form of a black powder which is odourless. It is soluble in water. It is a mixture of synthetic black dyes made by heating a mixture of nitrobenzene, aniline, and hydrochloric acid in the presence of a copper or iron. Nigrosin is stable if stored according to the recommended conditions. If exposed to excessive heat or in case of burning, it can generate hazardous decomposition products like nitrogen oxides, carbon monoxide, carbon dioxide and sulphur oxides. It is incompatible with strong acids and strong bases.

Nigrosin is widely used in industries as a colorant for lacquers and varnishes and in marker-pen inks. Nigrosin is also used for negative staining of bacteria and the capsule-containing fungus Cryptococcus neoformans. The shape and size of the organisms are seen as colour-free outlines against the dark background. One of the advantages of using nigrosin is that prior fixation by heat or alcohol is not needed, so the organisms are seen in more lifelike shapes. Furthermore, negative staining with nigrosin can reveal some microorganisms that cannot be stained by regular methods. Nigrosin WS is used in tests for viability because it enters dead cells while living cells exclude the dye.
