Proteinase K is a powerful enzyme which comes under the class of serine proteases. It is naturally obtained from the organism Tritirachium album and has the ability to digest a wide range of proteins. It is an endopeptidase and it shows a broad and generalized spectrum of activity against proteins. In molecular biology, it is a common reagent used during nucleic acid isolation and PCR. It degrades the proteins to allow the isolation of pure nucleic acids. It also has the ability to degrade and inactive nucleic acid degrading enzymes such as RNases and DNases, thus preserving the quality of nucleic acid isolated. The enzyme shows maximum activity in the pH range of 6.5 to 9.5 and is not deterred by the presence of other reagents used during nucleic acid preparations such as, SDS or EDTA. But the enzyme is rapidly inactivated in the presence of PMSF. The Proteinase K provided by Genei can be used by preparing a stock solution of 20 mg/mL in deionized water. The working concentration can be 50 g/mL (maintain temperature at 50 to 60C). It has an activity of 33.79 units/mg of protein, where, 1 unit of the enzyme can generate 1 mmol of Folin -positive amino acid in a minute at a temperature of 37 C.
