Tryptophan Medium is supplied by TM Media. APHA recommends the use of Tryptophan Broth to detect indole production by coliforms, a crucial characteristic for differentiating bacteria. This method determines whether certain bacteria can break down tryptophan into indole, which accumulates in the medium. Indole testing is useful in identifying microorganisms based on indole production, but further biochemical confirmation is necessary for complete identification. Tryptophanase is the enzyme that helps certain microorganisms break down tryptophan, which then produces indole through hydrolytic activity. The production of indole can be detected using Kovac's or Ehrlich's reagent, which combines with the aldehyde present to create a red colour in the alcohol layer. This red colour complex is then extracted and concentrated in the alcohol layer.
