Tth DNA Ligase ,250 U supplied by BLIRT is an enzyme isolated from Escherichia coli strain containing plasmid carrying the Thermus thermophilus DNA ligase gene and is known to catalyze the NAD-dependent phosphodiester bonds formation between adjacent 3-hydroxyl and 5-phosphate termini in double stranded DNA. Tth DNA Ligase has proven to be stable and active in optimum ligation temperature that ranges between 45 65C (7 10C higher than that of T4 DNA ligase). Higher ligation temperature removes nonspecific ligation and this temperature is determined by the Tm of the substrates.
