Urea Indole Medium is supplied by TM Media. Urea Indole Medium is utilized to recognize Enterobacteria based on their ability to produce urease and indole and to trans-deaminate tryptophan. This medium is particularly beneficial in differentiating Proteus species from Salmonella and Shigella species. It is important to observe the results for urease production before the indole reaction, as the addition of Kovacs reagent can cause the medium to lose colour due to a drop in pH. L-Tryptophan, an important amino acid, is converted into skatole and indole, which can be detected using Kovacs Reagent (R008). Sodium chloride is included to maintain the osmotic balance, while phosphates assist in buffering the medium. Microbes that possess the enzyme urease break down urea and release ammonia, which is detected by the pH indicator phenol red. The resulting increase in alkalinity imparts a pink hue to the medium.
