It is specific for the phosphate groups of DNA and attaches itself to where there are high amounts of adenine-thymine bonding. It is mainly used for staining peripheral blood smears and specimens obtained from the bone marrow. When blood films are stained using Giemsa Stain, the nucleus and cytoplasm of white blood cells take on characteristic blue or pink coloration. The use of purified eosin and thiazine dyes minimizes lot-to-lot variation.
