DNA Electrophoresis is used toseparate and visualize DNA in a sample. The principle involves the migration of charged DNA through a gel medium, generally Agarose, under an electric field. Negatively charged DNA migrates towards the positive electrode, at a speed depending on its size and charge. When the tracking dye has traveledatleast 3/4th of the gel, current is stopped, and the gel is visualized under UV transilluminator. Due to staining of DNA sample with an intercalating dye such as ethidium bromide, the DNA bands of separation can be visualized under UV, and an approximate size of DNA can be determined by referring to the standard DNA ladder.
