This kit simplifies isolation of plasmid DNA from yeast by the spin-column procedure. In the procedure, the cell wall of yeast cells is lysed enzymatically with lyticase/zymolyase in presence of a buffer. Spheroplasts are then collected by centrifugation and subjected to alkaline lysis, after which it is applied on to the silica column for binding of the DNA molecules in the presence of high salt concentration. The adsorbed DNA is washed to remove contaminants, and the pure plasmid DNA is eluted in Elution Buffer. The plasmid yields depend on copy number, yeast strain and growth conditions. The purified plasmid DNA can be used in all molecular biology procedures such as digestion with restriction enzymes, cloning, PCR, transfection, in vitro translation, blotting and sequencing.
