Lyophilized Proteinase K 100mg-TPRK-100 is supplied Transiom Genomics. Proteinase K powder is a subtilisin-related serine protease that hydrolyzes a variety of peptide bonds and is frequently used to cleanup enzymatic reactions or cell lysates. Proteinase K is an endolytic protease that slices peptide bonds present at the carboxylic sides of aliphatic, aromatic or hydrophobic amino acids. A tetrapeptide is the smallest peptide to be hydrolyzed by this enzyme. Proteinase K cannot be inactivated by metal chelators, by thiol-reactive reagents or by specific trypsin and chymotrypsin inhibitors. Phenylmethylsulfonyl fluoride and diisopropyl phosphorofluoridate are used to completely inhibit the enzyme. It is advisable store the Proteinase K powder at 4ºC and the dissolved solution at -20ºC.
Some applications of this kit include, Isolation of genomic DNA from cultured cells and tissues. Removal of DNAses and RNAses when isolating DNA and RNA from tissues or cell lines. Determination of enzyme localization, Improving cloning efficiency of PCR products.
