It is isolated from the microorganism Staphylococcus aureus and is found to have high affinity for mammalian IgG. Matrices for affinity chromatography use biologically active ligands, such as protein A, which are cross linked to agarose. There are three main methods by which this is done - triazine condensation reaction, epoxy and divinyl sulfone activation and cyanogen bromide method. Protein A - CL Agarose Pre-packed column by Genei contains Protein A immobilized with agarose in the form of a pre-swollen 50% suspension in an aqueous medium. The protein A used in the case in truncated, thus eliminating the albumin binding domains of the ligand. This renders it specific to the Fc region of IgG.
