This test is based on a lateral flow chromatographic immunoassay.

It detects and differentiates both IgG and IgM antibody.

The test has a conjugate pad containing the recombinant rK39 antigen conjugated with colloidal gold and rabbit IgG-gold conjugates.

It also has a nitrocellulose membrane strip two test lines (M and G lines) and a control line (C line).

The M line is pre-coated with monoclonal anti -human IgM for the detection of anti-L. donovani IgM, the G line is pre-coated with reagents for the detection of anti-L. donovani IgG, and the C line is pre-coated with goat anti-rabbit IgG.

The sample is dispensed into a sample well. It migrates due to capillary action.

In case of the presence of the antibody IgM, a burgundy colour appears on the M line, indicating a positive test result.

In case of the presence of the antibody IgG, a burgundy colour appears on the G line, indicating a positive test result.

It also contains an internal control C line which must develop a burgundy colour.
