Transport Swabs w/Transport Medium Amies W/Charcoal is supplied by TM media. Amies developed a modification of Stuart's transport medium by replacing glycerophosphate with an inorganic phosphate buffer and adding charcoal to create a higher percentage of positive results. The presence of sodium thioglycollate and small amounts of agar provides a reduced environment, while charcoal neutralizes toxic materials for sensitive pathogens like Neisseria gonorrhoeae. The medium also includes calcium, magnesium, potassium, and sodium salts to promote the survival of gonococcal cells and control bacterial cell permeability. Phosphates buffer the medium. Sterile cotton-tipped swabs or wooden sticks are used to collect specimens, and the swab should be pushed one-third of the depth of the medium. The swab is forced to the bottom of the medium when the cap is screwed down firmly. The medium should be kept cool during transport but not frozen.
