RNA Purifications and Extractions

RNA purification and extraction is the process of isolating RNA samples from biological extracts. There are many procedures for RNA extraction, the most common of which is Guanidinium thiocyanate-phenol-chloroform extraction. This is aliquidliquidextraction method which employs Guanidinium thiocyanate to denature proteins present in the sample. RNA partitions into the aqueous phase of the mixture. The phases are produced by phenol and chloroform in the solution.

However, the process is complicated by the universal presence of ribonuclease enzymes in cells and tissues, which rapidly degrade RNA. For this reason, equipment used in RNA extraction procedures is thoroughly cleaned and treated with harsh chemicals to destroy RNases. Another method of RNA extraction in liquid nitrogen, using a mortar and pestle (or tissue pulverizers) is also efficient in preventing ribonuclease activity.
