MIU Medium is supplied by TM Media. The term MIU refers to Motility Indole Urea, a test that is valuable for detecting gram-negative bacilli, specifically bacteria belonging to the Enterobacteriaceae family. This single-tube test employs three different assessments to differentiate the organisms based on their motility, urease activity, and ability to produce indole. Microbes tested in MIU agar exhibit either dispersed growth or cloudiness that spreads outward from the central inoculation line for motile organisms, whereas non-motile organisms show limited growth along the stab line. These microorganisms contain urease, an enzyme that breaks down urea and liberates ammonia and carbon dioxide. Ammonia reacts with water to produce ammonium carbonate, which is an alkaline compound that raises the pH of the testing medium. The presence of phenol red in the medium causes a colour change from yellow to pink-red in alkaline conditions. The enzyme tryptophanase breaks down tryptophan found in casein enzymatic hydrolysate, resulting in the production of indole, skatole, and indole acetic acid. Indole reacts with p-dimethyl amino benzaldehyde, which is present in Kovac's reagent, to create a reddish-violet quinoidal compound.
