Alkaline peptone water is used commonly in microbiological laboratories for the growth and enrichment of vibrio species from several samples such as water, food, etc. The inoculum is first grown in alkaline peptone water for improved growth of vibrio species. Further, it may be plated onto selective medium. The pH of alkaline peptone water is adjusted to be about 8.4, which is best suited for the growth of vibrio. The nitrogen source, carbon source and important nutrients for the medium is provided by animal tissue extracts. Sodium chloride is also added to the medium to act as an osmotic equalizer. The alkaline peptone powder is added to water and autoclaved. The vibrio inoculum is added to the media then. Two species of vibrio, Vibrio cholerae and Vibrio parahaemolyticus, can be grown using this medium.
