Electrophoresis, as the name suggests is a separation technique achieved under the influence of electric current. When DNA loaded in a gel is exposed to electric current flow, DNA,being negatively charged, migrates to the positive electrode, at a pace dependent on its size and charge. At the end of the electrophoretic run, the DNA appears as separated bands of varying intensity, when observed under UV transilluminator. Due to staining of DNA with an intercalating dye such as ethidium bromide, DNA fluoresces under UV exposure. For tracking the electrophoretic run, a tracking dye is mixed with DNA sample, which is visible to the naked eye. A gel loading dye is also mixed to visualize the DNA sample while loading. Other biochemicals such as a buffer, DNA ladders, dyes, gels and solidifying agents like agar or acrylamide, kits etc. are required during the technique of DNA electrophoresis.
